ABSTRACT

Objective:To explore the influence of different mental status (evaluated by Likert scale) on the first success rate, image artifacts and examination time of cone beam CT (CBCT) in infants and young children.Methods:Prospective analysis of children aged 1-6 years (98 cases) who underwent CBCT examination, before the examination, the children were scored in seven aspects: activity, adaptability, attention, mood, gender, age and parents′ attitude, and the total score was calculated, The total score was used as the test variable, and the success rate of the first examination, the artifact of the image and whether the examination time was longer than 2 minutes were used as the state variable. Using SPSS 16.0 to draw the ROC curve, the area under curve (AUC), significance, maximum Yoden′s index and their corresponding diagnostic (evaluation) critical values were obtained.Results:The success rate of the first examination was 78.6%(77/98), the AUC was 0.949 ± 0.025, the significance P < 0.05, the maximum Yoden′s index was 0.766, the corresponding critical value of diagnosis (evaluation) was 15; the artifact-free rate was 67.3%(66/98), the AUC was 0.873±0.038, the significance P < 0.05, the maximum Yoden′s index was 0.549, and the corresponding critical value of diagnosis (evaluation) was 15; the proportion of examination time in 2 minutes was 80.6%(79/98), the AUC was 0.854 ± 0.039, the significant P < 0.05, the maximum Yoden′s index was 0.614, and the corresponding critical value of diagnosis (evaluation) was 14. Conclusions:The Likert scale scoring method based on Thomas′s temperament analysis theory has certain application value in the psychological evaluation of children before cone-beam CT examination. For children whose total score is greater than or equal to 14, sedation or psychological counseling should be carried out before examination in order to effectively improve the success rate of examination and reduce the occurrence rate of artifacts in images, further more to reduce radiation dosage and increase the examination circulation of CBCT in children.

Methods 1752 patients aged 1 to 6 were selected for this retrospective study. They were divided into sedated group (219 cases) and non-sedated group (1 533 cases). The success rate and image quality were compared between two groups. Results The sedated group had a higher success rate to non-sedated group: 99.5%(218/219) vs. 90.4% (1 386/1 533). The motion artifact in sedated group was lower than non-sedated group with I degree: 4.8% (15/314) vs. 20.1%(327/1 630) and II degree: 0.3%(1/314) vs. 12.2%(199/1 630). Conclusion Giving chloral hydrate to pediatric patients before their CBCT exam would improve both success rate and image quality, and reduce unnecessary radiation expose.

ABSTRACT

Objective To investigate the expression and clinical significance of serum intercellular adhesion molecule-1(ICAM-1),soluble intercellular adhesion molecule-1(sICAM-1)and high sensitive C-reactive pro-tein(hs-CRP)in the patients with plasma cell mastitis(PCM).Methods 45 cases of PCM and 45 persons un-dergoing healthy physical examination in the Zaozhuang Municipal Maternal and Child Health Care Hospital from August 2015 to February 2017 were selected and served as the PCM group and control group.The ex-pression levels of serum CAM-1,sICAM-1 and hs-CRP were detected in the two groups.Then the results were statistically analyzed.Results The vascular endothelial cell ICAM-1 level in the PCM group was slightly low-er than that in the control group,while the ductal endothelial cell ICAM-1 level was higher than that in the control group,the difference was statistically significant(P<0.05);the levels of serum sICAM-1 and hs-CRP levels in the PCM group were significantly higher than those in the control group,the difference was statisti-cally significant(P<0.05).The ductal endothelial cell ICAM-1,sICAM-1 and hs-CRP levels after treatment in the PCM group were significantly lower than those before treatment,the difference was statistically significant (P<0.05).Conclusion The expression of serum ICAM-1,sICAM-1 and hs-CRP is closely correlated with PCM disease,can serve as the effective indicators for its clinical judgment and is worth promotion and applica-tion.

Objective:To clone swine leukocyte antigen,class II,DO alpha (SLA-DOA) gene from Banna mini-pig inbred line (BMI) and detect its mRNA expression level in 19 important tissues.Methods:The complete eDNA sequence of SLA-DOA gene was cloned by RT-PCR method from BMI and the mRNA expression pattern in BMI important tissues was examined by semi-quantative RT-PCR method.Nucleotide and protein sequences of SIA-DOA were used to carry out bioinformatics analysis and construct the phylogenetic tree.Results:The eDNA length of BMI SLA-DOA was 1 079 bp,which encoded a protein of 250 amino acids with molecular weight (Mw) 27.81 kD,and isoelectric point (pI) 6.48.Genome structure analysis showed it localized to Sus scrofa chromosomes 7 and consisted of four exons and three introns.Semi-quantitative expression analysis showed that SLA-DOA gene expressed highly in the lymph nodes and stomach;weakly in the heart,skin and duodenum and none in other 14 tissues.Functional bioinformatics analysis indicated that SLA-DOA protein contained one signal poptide,one transmembrane region,three conserved domains,four O-GlcNAc glycosylation sites,18 potential phosphorylation sites and to be located in the cytoplasm with 94.1% certainty.Phylogenetic analysis demonstrated that BMI (pig) had the closest relationship with cattle.Conclusion:This study have successfully cloned the SLA-DOA gene of Banna mini-pig inbred line,performed bioinformatics analysis and tissue expression profile analysis.It will provide a basis for the studies of BMI xenotransplantation.

ABSTRACT

Objective To observe the effect of the optimized formulas of extracts of Radix Astragali and Radix Angelicae Sinensis on the survival status of the idiopathic pulmonary fibrosis (IPF) mice,and on the expression levels of transforming growth factor-β (TGF-β) and vascular endothelial growth factor(VEGF),so as to optimize the therapeutic regimen and to explore the therapeutic mechanism.Methods One hundred and five SPF ICR male mice were randomly divided into normal group,model group and 5 Chinese medicine treatment groups (group 1,2,3,4,5 of the optimized formula of Radix Astragali and Radix Angelicae Sinensis extracts).The mice in the model group and the 5 treatment groups were intratracheally injected with bleomycin (5 mg/kg) to induce the pulmonary fibrosis model.On day 21,the lung tissues were taken out for the test.Hydroxyproline content was detected by alkaline hydrolysis method,and morphological changes of lung tissues were observed by hematoxylineosin (HE) staining and Mallory's staining methods.The expression levels of TGF-β and VEGF were detected by polymerase chain reaction (PCR).Results The HE staining and Mallory's staining results showed that the pulmonary fibrosis in the 5 treatment groups was relieved as compared with that in the model group,especially in the group 1,and the alveolar structure recovered better.The 21-day overall death rate in the treatment groups were lower than those in the model group,and group 1 and group 5 had the lowest rates,the difference being statistically significant (P< 0.05).Compared with the model group,the content of hydroxyproline in the lung tissues of the treatment groups were decreased to some degrees,and there was significant difference (P < 0.05 or P < 0.01).The expression levels of TGF-β and VEGF in model group were higher than those in normal group,but were deceased in the treatment groups to some degrees,except TGF-β expression in group 5,and the difference was significant(P < 0.05 or P < 0.01).Conclusion When the contents of Radix Astragali water-extract and Radix Angelicae Sinensis alcohol-extract were predominated,the extract formula exerts certain effects on decreasing hydroxyproline content in the lung tissues,inhibiting the expression levels of TGF-β and VEGF,and relieving the degree of pulmonary fibrosis in IPF mice.

ABSTRACT

Aim To establish an in vitro early drug cardiac tox-icity evaluation method by human embryonic stem cells derived cardiomyocytes ( hESC-CM) and real-time cell analysis Cardio (RTCA Cardio) system. Method The hESC-CM were cultured at RTCA Cardio E-Plate 96. Impedance signals from hESC-CM were analyzed for beating rate, contraction amplitude and beating rhythm irregularity to determine the optimum inoculation density and detection duration. Based on this, we used 0. 1 % DMSO to be the solvent and quinidine (0. 2, 0. 78, 3. 13, 12. 5, 50 and 100 μmol·L - 1 ) known as affecting cardiac activity to validate this method. Result The results revealed no significant changes in the cell index (CI), transient pulse patterns, beating rate and amplitude of hESC-CM. Quinidine will affect the CI and transi-ent pulse patterns of hESC-CM and decrease the beating rate and amplitude of hESC-CM when its concentration ≥3. 13 μmol · L - 1 . And this effect is concentration-dependent, the higher the concentration,the more time they need to recover beating and the more significant the beating rate and amplitude inhibition of quinidine on hESC-CM. Conclusion The method established by hESC-CM and RTCA Cardio system can detect the effect of quinidine on the contraction of hESC-CM, and this indicates that this method has the potential to be an attractive high-throughput tool for screening potential drugs in early evaluation of drug car-diotoxicity.

ABSTRACT

Objective To explore the relationship between the fibroblast growth factor 2 (FGFR2) gene polymorphism (rs 2981582 ,rs 1219648 ,rs 2420946) and the breast cancer risk in Tibetan population ,Qinghai province .Methods This is a case con‐trol study .Peripheral blood samples from 210 breast cancer patients and 230 healthy women in Qinghai area were collected .DNA samples were extracted from peripheral blood cells .FGFR2 gene polymorphism (rs 2981582 ,rs 1219648 ,rs 2420946) were typed by Taqman‐MGB probe based on PCR and DNA sequencing ,then analyzed its correlation with breast cancer in Tibetan population , Qinghai province .Results The genotype frequencies of rs 2981582 CC ,CT and TT were 40 .48% ,39 .05% and 20 .47% among the breast cancer patients while 36 .09% ,48 .69% and 15 .22% among the controls .The genotype frequencies of rs 1219648 GG ,AG and AA were 24 .76% ,26 .19 % and 49 .05% among the patients while 23 .91% ,47 .39% and 28 .70% among the controls .The genotype frequencies of rs 2420946 CC ,CT and TT were 29 .05% ,45 .24% and 25 .71% among the patients while 30 .87% , 51 .74% and 17 .39% among the controls .The genotype frequencies of all genetic loci had no significant difference between rs 2981582 and rs 2420946 (P>0 .05) .But the genotype frequencies of rs 1219648 AA have statistical sense (P< 0 .05) ,compared with GG ,the incidence of breast cancer was remarkably increased with AA [OR=1 .65 ,95% CI= (1 .01 ,2 .69)] .Conclusion This study shows that FGFR2 rs1219648 AA is related to breast cancer risk among Tibetan population .

ABSTRACT

OBJECTlVE To establish a real-time cell analysis system ( RTCA) for early drug car-diotoxicity evaluation. METHODS An in vitro drug cardiotoxicity evaluation method was established using RTCA Cardio system and primary cultured cardiomyocytes of neonatal rats. The beating rate, am-plitude and beating rhythm irregularity ( BRl ) of cardiomyocytes were observered after antiarrhythmic drugs, such as quinidine and lidocaine were added, to assess the effect of the above method on cardio-toxicity evaluation. RESULTS RTCA Cardo E-Plate 96 was inoculated with primary cultured cardiomyo-cytes that began to beat after 24 h and beat regularly after 48 h. The stable beating was maintained for a minimum of three days. The beating of cardiomyocytes decreased rapidly from 155±5 to 0 after incuba-tion with quinidine. The beating recovered gradually after 6 h. Quinidine at 3.1μmol·L-1 caused the beat-ing rate to return to 124±16. Quinidine allowed the beating rate to return to normal when the concentra-tion was less than 100.0μmol·L-1 . The beating rate, amplitude and BRl of cardiomyocytes changed in a concentration-dependent manner when incubating with lidocaine. The higher the concentration, the more significant the inhibition of lidocaine on cardiomyocytes. CONCLUSlON The cardiotoxicity of quinidine and lidocaine can be detected accurately using RTCA Cardio system, suggesting that this system can be used in early evaluation of drug cardiotoxicity.

Objective To get TDRP1 gene of sterile and fertile boar of the Banna minipig inbred line (BMI), predict its function by bioinformatics analysis, and detect its expression patterns in the fertile boar.Methods Based on the NM_001198925 sequence, we designed specific primers and amplified BMI TDRP1 using RT-PCR method for sequen-cing and bioinformatics analysis.Meanwhile, the expression of TDRP1 in 17 organ tissues ( heart, liver, spleen, lung, kidney, thymus, lymph nodes, skin, duodenum, stomach, cerebrum, cerebellum, testis, epididymis, seminal vesicle, prostate, and bulbourethral gland) of fertile BMI boar and in the testis of sterile and fertile BMI boars was analyzed by semi-quantitative RT-PCR.Results The experiment obtained 680 bp cDNA sequence ( GenBank accession number:KJ186786) of BMI TDRP1, which encodes a protein of 186 amino acids with a predicted molecular weight (Mw) of 20.49 kDa and isoelectric point (pI) 5.86, and no signal peptide.It was a nuclear protein with a probability of 94.1%and had a leucine-rich nuclear export signals.Homology analysis of protein sequences revealed that BMI TDRP1 showed high identi-ty with that of humans, macaca mulatta, mouse and rat.The RT-PCR analysis showed that TDRP1 had a similar expression in the testes of sterile and fertile BMI boars.It was highly abundant in the seminal vesicle and prostate, moderately ex-pressed in cerebellum and testis and weakly expressed in cerebrum and kidney, while undetected in other 11 organ tissues. Conclusions We have cloned TDRP1 complete coding sequence, and found 2 SNPs,showing no difference in sequences and the testis mRNA expression levels between the fertile and sterile BMI boars.The multi-tissue transcription profile shows different expression levels in different organ tissues, being high in the seminal vesicle and prostate.The results of this study provide a foundation for further insight into the role of this gene in spermatogenesis.

ABSTRACT

BACKGROUND:With the development and improvement of liver transplantation technology, patients waiting for or undergoing liver transplantation have continued to increase in number. Due to their fears and concerns about post-transplantation rehabilitation, patients are under physical and mental stress, most of whom are shown to have a variety of mental disorders that affect rehabilitation. At present, mental problems of liver transplant patients are more concentrated in post-transplantation research, while patients waiting for liver transplantation do not get enough mental assessment and intervention. OBJECTIVE:To investigate the suitable mode of cognitive-behavioral therapy for liver transplant recipients waiting for donor liver and to evaluate the effect to improve patient’s physical and mental state. METHODS:Twelve liver transplant patients who underwent pre-transplantation assessment were randomly divided into experimental group and control group, each group with six cases. The control group received conventional treatment. The experimental group received cognitive-behavioral therapy in addition to conventional treatment. Self-rating anxiety scale (SAS) and vital signs were measured in the two groups on admission, 1 week and 3 weeks after admission. RESULTS AND CONCLUSION:The SAS score of the experimental group was lower than that of the control group at 1 and 3 weeks after admission, and SAS score decreased as the intervention extended. The systolic blood pressure of the experimental group was lower than that of the control group at 3 weeks after admission, but there was no significant difference in diastolic blood pressure between two groups. The heart rate of the experimental group was lower than that of the control group at 1 and 3 weeks after admission. The respiratory rate of the experimental group was lower than that of the control group at 3 weeks after admission. Cognitive-behavioral therapy can reduce the level of anxiety and keep vital signs stable with good feasibility and effectiveness in patients waiting for liver transplantation.

Objective:To compare the effect of three different titanium alloy surface topographies on the biological character fibro-blasts.Methods:Three different titanium alloy Ti6Al4V surface topographies were created by machining(M),direct current electro-chemical etching(DC)and alternating current electrochemical etching(AC)respectively,and standard sized samples were prepared. The surface topographies were observed field emission scanning electron microscopy(FE-SEM).Mouse L929 fibroblasts were cultured on different surfaces.The attachment of cells was examined by acridine orange staining and observed under fluorescent microscope.The proliferation of cells was measured using MTT assay.Results:Tubles with the diamete of 5 -20 μm were observed on the surface of the sample of group DC,and more evenly distributed tubles on that of group AC.Micro-sulci were observed and no microtuble was found on the sample surface of group M.L929 cells were attached on the surface of all the samples in the 3 groups.More adhered and better strached cells were observed on AC treated surface.The proliferation of fibroblasts on AC and DC surfaces was superior to that on M treated surface and at the 3 -5 day the proliferation of fibroblasts on the AC surface was higher than that on DC treated surface.Con-clusion:AC treated surface with the micro-submicro-nano hierarchical topography of titanium alloy has a superior biocompatibility and can promote the early attachment and proliferation of the fibroblasts.

Objective To evaluate the clinical effect of thrombolysis combined with prostaglandin El (PGE1) in the treatment of patients with acute pulmonary thromboembolism (APTE).Methods According to the digital table,88 patients with APTE corresponding to thrombolytic therapy were randomly divided into the simply thrombolytic group(49 cases) and the joint PGE1 group(39 cases).The simply thrombolytic group were given conventional thrombolysis,and the joint PGE1 group were given thrombolysis combined PGE1 therapy.The levels of PaO2,PaCO2,pulmonary artery systolic pressure(PAsP),cardiac troponin I(cTNI),B-type natriuretic peptide(BNP) were monitored,and the clinical efficacy was evaluated.Results After the treatment,the levels of PAsP,cTNI,BNP of the joint PGE1 group were better than those of the simply thrombolytic group(t =2.173,2.349,2.495,all P <0.05).The clinical effective rate of the joint PGE1 group was 82.1%,which was significantly higher than that of the simply thrombolytic group (59.2 %) (χ2 =5.339,P < 0.05).The critical event occurrence of the joint PGE 1 group was 7.7 %,which was significant lower than that of the simply thrombolytic group (24.5 %) (χ2 =4.333,P < 0.05).Conclusion Thrombolysis combined PGEI therapy may be more effective in reducing pulmonary artery pressure,protecting myocardia,improving the right heart function than only thrombolysis,and thus improve clinical symptoms and short-term prognosis in the APTE patients.

Objective To investigate the expression of visfatin mRNA in abdominal omental adipose tissue and its relationship with blood lipid in type 2 diabetes mellitus.Methods 161 type 2 diabetic patients were divided into two groups according to triglyceride ( TG ),high triglyceride group ( TG ≥ 1.7mmol/L) and normal triglyceride group(TG < 1.7 mmol/L).The expression of visfatin mRNA in abdominal omental adipose tissue was measured with Northern Blot.Visfatin plasma concentration,fasting plasma glucose (FPG),blood lipid profiles and other biochemical indicators were also measured.Results Comparing with normal triglyceride group,the diabetic patients in high triglyceride group had significantly increased levels of visfatin plasma concentration [ ( 129.07 ± 21.35)ng/mL vs ( 101.65 ± 15.23 ) ng/mL,t =2.295,P <0.05]and mRNA expression in omental adipose tissue( P <0.05).Visfatin plasma concentration was positively correlated with TG( β =0.592,P <0.05 ) and FPG( β =0.763,P <0.01 ).Visfatin mRNA expression in omental adipose tissue had no correlation with FPG,TG and other biochemical indicators.Conclusions The plasma visfatin level in T2DM patients may be associated with triglyceride metabolism.

ABSTRACT

Mesenchymal stem cells (MSCs) have received considerable attention for various therapeutic approaches in recent years. MSCs are also easy to genetically modify to express therapeutic genes by using lentiviral vectors. Because of the similarities in genetics, physiology and metabolism between non-human primates (NHPs) and humans, NHPs models are invaluable for researching human disorders and for developing therapeutic strategies. Therefore, MSCs derived from NHPs could be a powerful tool for cell therapy and genetic engineering. Studies from captive and free-ranging adult NHPs show that up to 100% were infected with simian foamy virus (SFV). In this study, we found that all cultured MSCs derived from adult cynomolgus monkey were infected with SFV by RT-PCR. Therefore, antiviral drugs must be added in MSCs culture. However, because of SFV infection and additive antiviral drugs, the infection efficiency of the lentiviral vectors reduced significantly. In this study, we improved the infection efficiency by disabled antiviral drugs before lentiviral infection. It might be provide technical assistance for the culture of adult cynomolgus monkey MSCs as genetically engineered cells applied to clinical and experimental research.

ABSTRACT

Objective To investigate the short-term outcomes and oncological safety of laparoscopy resection for colorectal cancer. Methods Between January 2004 and March 2009, 35 patients with colorectal cancer underwent laparoscopic-assisted surgery in our hospital, among which 32 underwent radical resection, 1 underwent laparoscopic exploration, and 2 were converted to open surgery. These patients were allocated in the laparoscopic group. Same numbers of patients who underwent conventional open surgery during the same period of time were identified and allocated in the open group. Short-term data, including surgical time, intra-operative blood loss, intra-operative blood transfusion, length of incision, histopathological data, post-operative complications and post-operative functions, were collected and compared between the two groups. Results A total of 67 patients were enrolled (32 in both laparoscopy and open groups). The other 3 cases were analyzed separately (1 underwent laparoscopic exploration and 2 converted to open surgery). The two groups were well balanced as to age, gender, and TNM staging and location of tumour. Histopathologically, the laparoscopic approach was equal to conventional approach as regard to resection margin (distal margin, 5 cm vs 5 cm, P=0.664) and lymph node yield (7 vs 8, P=0.228). This study also showed a longer surgical time (250 min vs 180 min, P=0.006), but shorter length of incision (10 cm vs 20 cm, P<0.001), less demand of intra-operative blood transfusion (1 case vs 10 cases, P=0.003), reduced use of analgesics (12 cases vs 25 cases, P=0.004), shorter post-operative stay (9.5 days vs 11 days, P=0.008) and earlier recovery of bowel function of the laparoscopic group. There was no statistical difference of intra-operative blood loss (200 mL vs 200 mL, P=0.098), incidence of post-operative complications (8 cases vs 6 cases, P=0.545) and volume of post-operative negative drainage (507.5 mL vs 669.0 mL, P=0.475) between the two groups. Conclusions Though limited by a relatively small sample size, our study showed that laparoscopy approach for colorectal cancer is equal to open approach in terms of oncological safety and short-term outcome.

ABSTRACT

An automatic detection of Electrocardiogram (ECG) using Quantum Neural Networks (QNN) with multiresolution analysis is given in the paper. QNN originates from exploiting BP neural networks. With the quantum neurons, QNN can model the levels of uncertainty arising from complex classification problems. Its potential advantage over conventional methods is based on the argument for reliability. The fuzzy feature is expected to enhance the reliability of the network, which is critical for improving desirable diagnosis accuracy. And wavelet transform with multiresolution analysis is adopted in ECG pretreatment for reducing the numbers of neurons. So it can improve convergence speed of the network. The results of simulation confirmed the feasibility of the proposed approach.

ABSTRACT

Objective To assess the sensitivity of indices of hip development in children with spastic diple-gia resulting from cerebral palsy. Methods X-ray images of the hips of 57 children with cerebral palsy ( the cere-bral palsy group) were checked, and the acetabular index ( AI), femur head migration percentage ( MP), center-edge angle and neck-shaft angle (NSA) were compared with those of normal children ( the control group, n = 30).Results The differences in MP and NSA between the two groups were significant. The prevalence of hip subluxation was 20.45% among the children with spastic diplegia who could not walk independently, and the prevalence was sig-nificantly greater in children 3 to 5 years old than among those under 3. Conclusion The MP can be used as a sen-sitive index to evaluate hip development. Age is a relevant factor affecting the hip development of children with cere-bral palsy.

ABSTRACT

Objective To study the clincial effects of of single Sialic four hexose sodium injection ganglio-side (GM1) in treating periventricular leukomalacia of premature infants. Methods 41 cases of periventricular ]eukomalacia patients were divided into GMI treatment group (n=22) and control group (n=19). The treatment group were given another GM1 treatment. Dynamic observation of changes in B-head information was conducted. Re-stilts 6 months after birth, 8 cases(36.4%) in treatment group and 13cases (68.4%)in control group occurred with ventricular dilatation (χ28.85, P<0.05). Nervous system examination revealed there were 5 cases (22.7%) in treatment group and 10 cases (52.6%) in control group with limited performance of the Board of mus-cle tone and abnormal movements (χ2 3.93, P<0.05); 4 cases in treatment group (18.2%) and 9 cases (47.4%) of control group had mild cognitive impairment and mental retardation according to nervous system sequel-ae(χ24.01,P<0.05). Conclusion GM1 can effectively treat periventricular leukomalacia and reduce abnormal movements.
